TL DR version of what Enduro & Waste said: The treasure mission won't be offered again, so, without using cheats/exploits, you can only get into the Treasure Room twice (per playthrough). Once you turn in that mission, there is a second treasure mission that just repeats the last part of the story, starting at the lighthouse, and you can fight the leviathan and get back in the Treasure Room a second time that way.
